Shelton Jackson "Spike" Lee (d. 20 Mart 1957; Atlanta, Georgia), Amerikalı oyuncu, yönetmen, film yapımcısı, senarist ve akademisyen.

Emmy Ödüllü ve iki Akademi Ödülü adaylığı olan Spike Lee'nin filmleri genelde sosyal veya politik içeriklidir. Lee ayrıca öğretmendir. Aynı zamanda koyu bir New York Knicks taraftarıdır. Knicks'in her maçında Lee'yi izleyiciler arasında görmek mümkündür.
